Innovations in lightmicroscopy have revolutionized the way researchers study biological systems.Although fluorescence microscopy is currently the method of choice for cellularimaging, it faces several fundamental limitations such as the rather bulky fluorescenttags, color barrier for multiplex imaging, and limited ability for probing invivo metabolism. To address these challenges, I will present three chemicalimaging strategies, respectively. First, we devised a Bioorthogonal ChemicalImaging platform suited for probing the dynamics of small bio-molecules, whichcannot be effectively labeled by bulky fluorophores. This scheme couples theemerging stimulated Raman scattering microscopy with tiny and Raman-activevibrational probes (e.g., alkynes and stable isotopes). Exciting biomedicalapplications such as imaging fatty acid metabolism related to lipotoxicity,glucose uptake and metabolism, drug trafficking, protein synthesis in brain,DNA replication, protein degradation, RNA synthesis and tumor metabolism willbe presented. Second, we invented a super-multiplex optical imaging technique.We developed electronic pre-resonance stimulated Raman scattering (epr-SRS)microscopy, achieving exquisite vibrational selectivity with high versatilityand sensitivity. Chemically, we created a unique vibrational palette consistingof novel dyes, each displaying a single epr-SRS peak in the cell-silent spectrawindow. Up to 24 resolvable colors are currently achieved. This super-multiplexoptical imaging approach promises to facilitate untangling the intricateinteractions in complex biological systems, and can also find broadapplications in photonics and biotechnology in general. Third, we introduced aplatform that combines deuterium oxide (D2O) probing with SRS microscopy(DO-SRS) to image in situ metabolic activities in animals including C. elegans,zebrafish and mice. Enzymatic incorporation of D2O-derived deuterium intomacromolecules generates carbon-deuterium (C-D) bonds, which track biosynthesisin tissues in situ. Within the broad vibrational spectra of C-D bonds, wediscovered lipid-, protein-, and DNA-specific Raman shifts with macromolecularselectivity. DO-SRS enabled us to probe de novo lipogenesis in animals, imageprotein biosynthesis without tissue bias, and simultaneously visualize lipidand protein metabolism. DO-SRS, being noninvasive, universally applicable, andcost-effective, can be adapted to a broad range of biological systems to studydevelopment, tissue homeostasis, aging, and tumor heterogeneity.  

闵玮教授个人简历: 

闵玮教授2003年本科毕业于北京大学,2008年博士毕业于哈佛大学,师从美国科学院院士谢晓亮教授,之后继续在谢教授团队从事博士后工作,2010年起执教哥伦比亚大学,2016年被授予终身教职,2017年起任职正教授。闵教授课题组的研究方向是基于分子光谱学开发新型光学显微镜,结合化学探针和生物技术,推动神经科学、癌症检测和疾病诊断等前沿生命科学和医学课题的发展。闵玮教授已经在Science,Nature, Nature Methods, PNAS等顶级刊物上发表论文多篇。其中,闵玮教授团队首次使用受激拉曼散射显微镜突破传统光学成像的颜色极限的工作曾被多个顶级期刊如Nature,Nature Methods专题报导。 

Dr. Wei Min graduatedfrom Peking University with a Bachelor's degree of Chemistry in 2003.  He received his Ph.D. from Harvard Universityin 2008 studying single-molecule biophysics with Prof. Sunney Xie. Aftercontinuing his postdoctoral work in Xie group, Dr. Min joined the faculty ofDepartment of Chemistry at Columbia University in 2010, and is promoted to FullProfessor there in 2017. He is also affiliated with the Kavli Institute forBrain Science at Columbia University. 

Dr. Min's currentresearch interests focus on developing novel optical spectroscopy andmicroscopy technology to address biomedical problems. In particular, his grouphas made important contributions to the development of stimulated Ramanscattering (SRS) microscopy and its broad application in biomedical imagingincluding bioorthogonal chemical imaging of small molecules and super-multiplexvibrational imaging.Dr. Min’s work has beenpublished by Science, Nature, Nature Methods, PNAS, Nature Communications etc.;and has been reported by many major journals such as Nature, Nature methodsetc.. 

Dr. Min's contribution has been recognized by a number of honors,including Analyst Emerging Investigator Lectureship (2018), Coblentz Award ofMolecular Spectroscopy (2017), the ACS Early Career Award in ExperimentalPhysical Chemistry (2017), Buck-Whitney Award of American Chemical Society(2015), Camille Dreyfus Teacher-Scholar Award (2015), George Fraenkel FundAward (2014), Alfred P. Sloan Research Fellowship (2013), NIH Director's NewInnovator Award (2012) and Faculty Finalist of Blavatnik Awards for YoungScientists of the New York Academy of Sciences (2012).
